Methods for increasing the production of ethanol from microbial fermentation
A stable continuous method for producing ethanol from the anaerobic bacterial fermentation of a gaseous substrate containing at least one reducing gas involves culturing in a fermentation bioreactor anaerobic, acetogenic bacteria in a liquid nutrient medium; supplying the gaseous substrate to the bioreactor; and manipulating the bacteria in the bioreactor by reducing the redox potential, or increasing the NAD(P)H TO NAD(P) ratio, in the fermentation broth after the bacteria achieves a steady state and stable cell concentration in the bioreactor. The free acetic acid concentration in the bioreactor is maintained at less than 5 g/L free acid. This method allows ethanol to be produced in the fermentation broth in the bioreactor at a productivity of greater than 10 g/L per day. Both ethanol and acetate are produced in a ratio of ethanol to acetate ranging from 1:1 to 20:1.

Process for production of alcohols by microbial fermentation
The invention provides method for producing butanediol and specifically 2,3-butanediol by anaerobic fermentation of a gaseous substrate comprising CO by a culture of at least one micro-organism. In accordance with particular methods of the invention, 2,3-butanediol is produced by anaerobic fermentation of substrates including carbohydrates and carbon monoxide. The invention further provides for the up-regulation of a native 2,3-butanediol dehydrogenase gene in the at least one micro-organism. The 2,3-butanediol can be further converted to compounds such as butene(s), butadiene and methyl ethyl ketone. These compounds can be further converted to chemical products such as 2-butanol.

A method is provided for controlling a metabolic profile of an anaerobic microbial fermentation culture. In particular, a metabolic profile of a fermentation process is controlled by controlling the amount of dissolved CO2 provided to a culture. Further provided is a method of producing one or more products by microbial fermentation of a gaseous substrate through feeding tail gas CO2 from a reactor to a second reactor, or by recycling tail gas CO2 to the same reactor.

Ammonia-specific 5'-XMP aminase mutant
Disclosed herein are ammonia-specific 5'-XMP aminase mutants and a method for preparing the same. A mutation is introduced into the active site of glutamine-dependent catalysis in 5'-XMP aminase. The resulting 5'-XMP aminase mutant is devoid of the glutamine-dependent activity and specifically reacts with external ammonia in converting 5'-XMP into 5'-GMP. Thus, the ammonia-specific 5'-XMP aminase mutant is stabler within cells compared to the wild type, and can be useful in the industrial conversion of 5'-XMP into 5'-GMP.
